The Facts of Wash Buffer Containing RNA Samples
Lead to low A260/230. It will be more obvious when you have a low RNA concentration. After you normalize your RNA samples to the same concentration, the wash buffer in each concentration-normalized samples are not the same. The orignal low concentrated RNA samples will have more wash buffer in the concentration-normalized samples because you will add more wash buffer-containing-samples to get the same quantity. When you do reverse transcription, reverse transcriptase activity will be inhibitted. The more wash buffer in your samples the more inhibition of reverse transcriptase activities. Thus, there will be less cDNA in the orginal low RNA samples although you put same amount RNA in the reverse transcriptase reaction.

The final consequences will be: when you do quantitative real-time PCR you will get non-accurated results; when you clone genes, especially for those low expression genes, you will get very low PCR products or couldn’t get any PCR products.
